WIL Research to Demonstrate How to Shorten Discovery PK Turnaround at Thermo Scientific User Meeting

ASHLAND, Ohio--(BUSINESS WIRE)-- A WIL Research bioanalytical chemist, Dr. Hui Tao, will discuss how the Thermo Scientific Q-Exactive Quadruple Orbitrap High Resolution Mass Analyzer can accelerate discovery pharmacokinetic (PK) screening time by 20 percent at the 2012 Thermo Scientific Mass Spectrometry User Meeting.

One of the main challenges in discovery PK is the need for quick throughput. CROs capable of screening 100s of compounds per week are in demand. Those CROs that can achieve a low limit of quantification (e.g., 0.1 nM), wide dynamic ranges, and rapidly troubleshoot issues that occur will be particularly well suited for pharma partnerships.

WIL Research is the first CRO to use the Q-Exactive high-resolution mass analyzer to support discovery PK. Hui Tao says the Q-Exactive has saved WIL’s Discovery PK team considerable time and effort by allowing them to quickly move between quantitation and identification/resolution of compound/matrix issues. Hui comments that the instrument is very user friendly and automated.

“Chemists typically spend about a day per week on adjusting instrument settings to get good quantitation results for each compound being screened,” Tao said. “However, the Q-Exactive provides an easy process for compound tuning that saves us hours of instrument time.”

Case studies demonstrate power to quantitate analytes

The first case study Tao will present focuses on the identification and quantitation of five analytes in rat plasma using three internal standards under isocratic conditions. WIL used the Q-Exactive to quickly identify and quantitate co-eluting analytes and metabolites, which saved hours by reducing chromatography requirements.

Tao and the WIL team also investigated “ghost” peaks, matrix interference often associated with PK screening when using low-identification-power instruments. They were able to use the Q-Exactive to enhance the sensitivity and quantitation range by eliminating this matrix effect.

“The Q-Exactive has the ability to deliver highly sophisticated investigations to allow smooth and rapid transitions between troubleshooting and actual quantitation,” said Tao.

In addition to the Q-Exactive, WIL Research has several other platforms for performing discovery PK screening activities. Having different mass analyzers and ESI probes allows complementary platforms to be available, which is particularly important when performing difficult assays.

The Thermo Scientific Mass Spectrometry User Meeting is a chance for researchers to learn from fellow users through customer presentations, software workshops, successful quantitation, and characterization and quantitation of biologics.
